The rhizosphere is an active compartment where plant and microorganisms establish a molecular dialogue. In this study, we analysed the impact of Arabidopsis thaliana on bacterial community structure and the expression of certain beneficial genes using DNA- and mRNA-SIP in the rhizosphere of plantlets grown under (13)CO(2) for 13, 21 and 27 days. DNA- and rRNA-SIP revealed changes in bacterial communities inhabiting the rhizosphere soil that were probably related to modification of root exudates, while root-colonizing populations were maintained over time suggesting their metabolic versatility and adaptation. The impact of the plant via root exudates on the expression of the noncoding RNAs rsmZ, acdS gene encoding 1-aminocyclopropane-1-carboxylate deaminase and nosZ gene encoding nitrous oxide reductase, in the root-adhering soil and on the roots of A. thaliana was determined using mRNA-SIP. Results showed that these genes were present and expressed by bacteria inhabiting roots and by those that derive nutrients from the breakdown of organic matter in soils or from root exudates. The expression of rsmZ under natural conditions indicates the importance of noncoding RNAs in bacterial adaptation to their ecological niches. 相似文献

Background
Neutrophils play a major role in inflammation by releasing large amounts of ROS produced by NADPH-oxidase and myeloperoxidase (MPO). The proinflammatory cytokine TNFα primes ROS production through phosphorylation of the NADPH-oxidase subunit p47phox on Ser345. Conventional anti-inflammatory therapies remain partially successful and may have side effects. Therefore, regulation of neutrophil activation by natural dietary components represents an alternative therapeutic strategy in inflammatory diseases such as inflammatory bowel diseases. The aim of this study was to assess the effect of punicic acid, a conjugated linolenic fatty acid from pomegranate seed oil on TNFα-induced neutrophil hyperactivation in vitro and on colon inflammation in vivo.Methodology and Principal Findings
We analyzed the effect of punicic acid on TNFα-induced neutrophil upregulation of ROS production in vitro and on TNBS-induced rat colon inflammation. Results show that punicic acid inhibited TNFα-induced priming of ROS production in vitro while preserving formyl-methionyl-leucyl-phenylalanine (fMLP)-induced response. This effect was mediated by the inhibition of Ser345-p47phox phosphorylation and upstream kinase p38MAPK. Punicic acid also inhibited fMLP- and TNFα+fMLP-induced MPO extracellular release from neutrophils. In vivo experiments showed that punicic acid and pomegranate seed oil intake decreased neutrophil-activation and ROS/MPO-mediated tissue damage as measured by F2-isoprostane release and protected rats from TNBS-induced colon inflammation.Conclusions/Significance
These data show that punicic acid exerts a potent anti-inflammatory effect through inhibition of TNFα-induced priming of NADPH oxidase by targeting the p38MAPKinase/Ser345-p47phox-axis and MPO release. Pinus pinaster (Ait.) somatic embryogenesis (SE) has been developed during the last decade, and its application in tree improvement programs
is underway. Nevertheless, a few more or less important problems still exist, which have an impact on the efficiency of specific
SE stages. One phenomenon, which had been observed in embryogenic tissue (embryonal mass, EM) initiated from immature seed,
has been the loss of the ability to produce mature somatic embryos after the tissue had been cultured for several months.
In an attempt to get insight into the differences between young cultures of EM (3-mo-old since the first subculture) of P. pinaster that produced mature somatic embryos and the same lines of significantly increased age (18-mo-old, aged EM) that stopped
producing mature somatic embryos, we analyzed in both types of materials the levels of endogenous hormones, polyamines, the
global DNA methylation, and associated methylation patterns. In addition, we included in the analysis secondary EM induced
from mature somatic embryos. The analysis showed that the two tested genotypes displayed inconsistent hormonal and polyamine
profiles in EM cultures of a similar phenotype and that it might be difficult to attribute one specific profile to a specific
culture phenotype among genotypes. Experiments were also undertaken to determine if the global DNA methylation and/or the
resulting methylation pattern could be manipulated by treatment of the cultures with a hypomethylating drug 5-azacytidine
(5-azaC). An aged EM was exposed to different concentrations and durations of 5-azaC, and its response in culture was established
by fresh mass increases and somatic embryo maturation potential. All of the analyses are new in maritime pine, and thus, they
provide the first data on the biochemistry of EM in this species related to embryogenic potential. 相似文献

A literature survey identified 403 primary research publications that investigated the ecological effects of invasive alien
insects and/or the mechanisms underlying these effects. The majority of these studies were published in the last 8 years and
nearly two-thirds were carried out in North America. These publications concerned 72 invasive insect species, of which two
ant species, Solenopsis invicta and Linepithema humile, accounted for 18% and 14% of the studies, respectively. Most publications investigated effects on native biodiversity at
population or community level. Genetic effects and, to a lesser extent, effects on ecosystem services and processes were rarely
explored. We review the effects caused by different insect invaders according to: their ecosystem roles, i.e. herbivores,
predators, parasites, parasitoids and pollinators; the level of biological organisation at which they occur; and the direct
and indirect mechanisms underlying these effects. The best documented effects occur in invasive ants, Eurasian forest herbivores
invasive in North America, and honeybees. Impacts may occur through simple trophic interactions such as herbivory, predation
or parasitism. Alien species may also affect native species and communities through more complex mechanisms such as competition
for resources, disease transmission, apparent competition, or pollination disruption, among others. Finally, some invasive
insects, particularly forest herbivores and ants, are known to affect ecosystem processes through cascading effects. We identify
biases and gaps in our knowledge of ecological effects of invasive insects and suggest further opportunities for research.

Labile soil carbon inputs mediate the soil microbial community composition and plant residue decomposition rates 总被引:2,自引:0,他引:2
Root carbon (C) inputs may regulate decomposition rates in soil, and in this study we ask: how do labile C inputs regulate decomposition of plant residues, and soil microbial communities? In a 14 d laboratory incubation, we added C compounds often found in root exudates in seven different concentrations (0, 0.7, 1.4, 3.6, 7.2, 14.4 and 21.7 mg C g(-1) soil) to soils amended with and without (13) C-labeled plant residue. We measured CO(2) respiration and shifts in relative fungal and bacterial rRNA gene copy numbers using quantitative polymerase chain reaction (qPCR). Increased labile C input enhanced total C respiration, but only addition of C at low concentrations (0.7 mg C g(-1)) stimulated plant residue decomposition (+2%). Intermediate concentrations (1.4, 3.6 mg C g(-1)) had no impact on plant residue decomposition, while greater concentrations of C (>7.2 mg C g(-1)) reduced decomposition (-50%). Concurrently, high exudate concentrations (>3.6 mg C g(-1)) increased fungal and bacterial gene copy numbers, whereas low exudate concentrations (<3.6 mg C g(-1)) increased metabolic activity rather than gene copy numbers. These results underscore that labile soil C inputs can regulate decomposition of more recalcitrant soil C by controlling the activity and relative abundance of fungi and bacteria. 相似文献

From necrotic tissue of a Nashi pear tree, 24 Erwinia pyrifoliae strains, found to be identical by pulsed-field gel electrophoresis analysis, were isolated. Thirteen strains were not virulent on immature pears and did not induce a hypersensitive response in tobacco leaves. It was recently shown that vascular endothelial growth factor (VEGF), a growth factor for endothelial cells, plays a pivotal role in rheumatoid arthritis. VEGF binds to specific receptors, known as VEGF-RI and VEGF-RII. We assessed the physical and histological effects of selective blockade of VEGF and its receptors in transgenic K/BxN mice, a model of rheumatoid arthritis very close to the human disease. Mice were treated with anti-mouse VEGF Ab, anti-mouse VEGF-RI and -RII Abs, and an inhibitor of VEGF-RI tyrosine kinase. Disease activity was monitored using clinical indexes and by histological examination. We found that synovial cells from arthritic joints express VEGF, VEGF-RI, and VEGF-RII. Treatment with anti-VEGF-RI strongly attenuated the disease throughout the study period, while anti-VEGF only transiently delayed disease onset. Treatment with anti-VEGF-RII had no effect. Anti-VEGF-RI reduced the intensity of clinical manifestations and, based on qualitative and semiquantitative histological analyses, prevented joint damage. Treatment with a VEGF-RI tyrosine kinase inhibitor almost abolished the disease. These results show that VEGF is a key factor in pannus development, acting through the VEGF-RI pathway. The observation that in vivo administration of specific inhibitors targeting the VEGF-RI pathway suppressed arthritis and prevented bone destruction opens up new possibilities for the treatment of rheumatoid arthritis. 相似文献
